Hygiena ZymoSnap™ ALP Positive Control Kit – 350 mU/L, 5 Vials

The Hygiena ZymoSnap ALP Positive Control Kit contains 5 vials of alkaline phosphatase (ALP) standard at 350 mU/L, the critical limit for pasteurization verification. You need it to set and check the pass/fail limits for Hygiena ZymoSnap ALP tests. ZymoSnap results in RLU vary with milk type and fat content, so each product you test needs its own fail limit, set by running this control in that same milk.

Each vial is reconstituted with a heat-treated negative control made from the milk you're testing, so the control matches your product. Hygiena lists expected ranges for 4%, 2% and 0.1% fat milk. The Hygiena ZymoSnap Positive Control Kit is available in Canada from Cambridge Environmental Products with CAD pricing and Canadian purchasing support.

Ideal For

  • Setting pass/fail RLU limits for each milk type tested with ZymoSnap ALP
  • Monthly verification of ZymoSnap ALP test performance
  • Checking each new lot of ZymoSnap ALP devices
  • Dairy QA/QC labs documenting test and luminometer performance

Why You Need It

Sets your fail limit at 350 mU/L

The control contains ALP at 350 mU/L. The average RLU of the positive control in a given milk becomes the fail limit for that product on your luminometer.

Matched to your product

You reconstitute each vial with a negative control made by heat-treating the same milk you test. The control then reflects the fat content and makeup of your own product.

Confirms the system is working

Running negative and positive controls when you start, monthly and with each new lot of devices confirms the tests, incubator and luminometer are all performing correctly.

How to Use

  1. Make the negative control: heat 10 mL of the milk you test at 72 °C for 10 minutes, cool quickly on ice, cover and refrigerate.
  2. Test the negative control: run 3 ZymoSnap ALP tests. The average must be below 20 RLU.
  3. Make the positive control: remove the vial's seal and cap, add 1.0 mL of the matching negative control, recap and shake vigorously for 60 seconds until dissolved.
  4. Test the positive control: run 3 ZymoSnap ALP tests and check the average against the expected range.
  5. Set the limit: enter the positive control average as the fail RLU for that milk type on your luminometer.

Steps are from the Hygiena ZymoSnap ALP Positive Control Kit instructions. Use a separate vial and negative control for each milk type.

Expected Positive Control Results

Milk Fat Min RLU Max RLU
4% 222 249
2% 315 385
0.1% 548 784

Average of 3 replicates, per Hygiena. If the average falls outside the range, repeat 3 tests from the same vial.

Selection & Handling Notes

  • Equipment not included: you'll also need ZymoSnap ALP tests, a Hygiena EnSURE Touch or EnSURE, a 72 °C water bath or incubator, a 37 °C incubator, an ice bath, and pipettes for 75 µL and 1.0 mL.
  • One milk type per vial. Reconstitute each vial only with negative control made from the milk type being tested.
  • Test the negative control first. It must average below 20 RLU before you prepare the positive control.
  • Keep samples chilled. Reactivated and microbial phosphatase can give high readings.
  • Short stability once mixed: use reconstituted controls within 7 days.
