Burkholderia Cepacia Selective Agar (BCSA) USP is an agar medium recommended for the selective and differential isolation of Burkholderia cepacia complex (Bcc). B.cepacia is an important pathogen in pulmonary infections in patients with cystic fibrosis, and BCSA was devised for isolation of the organism from respiratory secretions of patients to be tested under research laboratory conditions. In 1997, Henry et al. 1 formulated a highly selective medium to improve the speed and accuracy of B.cepacia complex isolation and to provide quantitative recovery. This product is formulated according to USP <60>. There is a choice of 3 sizes - 500g, 2kg, and 10kg.
PREPARATION: Mix 50.6 grams of the medium in one liter of purified water until evenly dispersed. Heat with repeated stirring until boiling to dissolve completely. Autoclave at 121°C for 15 minutes. Cool to between 45°C and 50°C. Aseptically add 10 mL of a filter-sterilized solution containing 10.0 mg of Gentamicin, 2.5 mg Vancomycin, and 600,000 units of Polymyxin B. Mix thoroughly by swirling, with minimal agitation. Distribute into sterile petri dishes.
