Hygiena ZymoSnap™ ALP Alkaline Phosphatase Test – Dairy Pasteurization Verification, 100 Tests

Hygiena ZymoSnap ALP is a rapid test that measures alkaline phosphatase (ALP) in milk and dairy products to verify pasteurization. ALP is an enzyme naturally present in raw milk and destroyed by proper pasteurization, so a low ALP result confirms the product was heat-treated correctly. ZymoSnap uses bioluminescence and gives a quantitative result in about 5 minutes on the Hygiena EnSURE Touch or EnSURE luminometer.

Each device is an all-in-one test: add 75 µL of sample, snap to release the reagent, incubate for 5 minutes at 37 °C and read. Hygiena states the method is AOAC Performance Tested Method℠ validated, and that it measures below the 350 mU/L critical limit for ALP. It's sold in boxes of 100 single-use tests. Hygiena ZymoSnap ALP is available in Canada from Cambridge Environmental Products with CAD pricing and Canadian purchasing support.

Ideal For

  • Dairy processors: verifying pasteurization of each batch at the plant
  • Fluid milk: whole, semi-skimmed and skimmed cow's milk, and goat's milk
  • Cream, flavoured milks, shakes and yogurts (per Hygiena)
  • Dairy QA/QC and contract testing labs that want results in minutes instead of hours
  • Sites already using a Hygiena EnSURE Touch or EnSURE luminometer

Why Choose ZymoSnap ALP?

Pasteurization results in about 5 minutes

The assay takes 5 minutes of incubation plus about 10 seconds to read. A problem with the pasteurizer can be caught and the product held before it leaves the plant.

Quantitative and sensitive

Light output (RLU) is directly proportional to ALP activity. Hygiena states the test measures below the 350 mU/L critical limit for ALP in cow's milk, and compares its performance to the ISO 11816 / IDF 155 fluorometric reference method.

AOAC validated

Hygiena states ZymoSnap ALP is validated under the AOAC Performance Tested Methods℠ program. The certificate is linked under Product Documents.

Simple, all-in-one device

There are no reagents to prepare. Pipette the sample into the device, activate it and incubate. Results are stored on the luminometer and can be trended in Hygiena's SureTrend software. Hygiena states throughput is more than 45 tests per hour.

What You Need

Item Notes
ZymoSnap ALP devices This product: 100 tests per box
Hygiena luminometer EnSURE Touch or EnSURE (not included)
37 °C incubator Dry block incubator or water bath at 37 ±1 °C (not included)
Pipette and tips 75 µL (not included)
Positive control ZymoSnap ALP Positive Control Kit (ZS-ALP-PC), sold separately, to set pass/fail limits

How to Use

  1. Let the device stand at room temperature (20–25 °C) for 10 minutes.
  2. Lift the Snap-Valve bulb out of the tube and pipette 75 µL of sample into the tube. Replace the bulb and flick the device so all the sample reaches the bottom.
  3. Break the Snap-Valve and squeeze the bulb twice to release the reagent.
  4. Incubate at 37 ±1 °C for 5 minutes. Turn on the luminometer and select the product program during incubation.
  5. Shake for 5 seconds, insert the device into the luminometer and read. Results take about 10 seconds. Read each device only once.

Steps are from the Hygiena ZymoSnap ALP instructions for use. Pass/fail RLU limits must be set for each milk type using the positive control kit, and checked monthly and with each new lot.

Common Applications

Application How ZymoSnap ALP Helps
Batch release Confirms pasteurization in minutes before product ships.
Pasteurizer checks Catches under-processing or raw milk contamination quickly.
Multiple milk types Separate pass/fail limits can be set for each fat level and product.
Records and trending Results stored on the luminometer and in SureTrend software.

Selection & Handling Notes

  • Positive control kit required. Pass/fail RLU limits depend on milk type and fat content. Set them with the ZymoSnap Positive Control Kit, then re-check monthly and with each new lot of devices.
  • Not for UHT or ambient-stable products. Hygiena states the test isn't suitable for long-life milk.
  • Keep samples chilled. Reactivated phosphatase and microbial phosphatase can give high readings.
  • Control incubation closely. RLU varies with incubation temperature and time, so keep to 37 ±1 °C and 5 minutes.
  • Light sensitive: store devices at 2–8 °C in their bag until use.
  • Regulatory use: check the method against the pasteurization verification requirements that apply to your facility.

Frequently Asked Questions

What does ZymoSnap ALP test for?

Alkaline phosphatase (ALP), an enzyme in raw milk that's destroyed by proper pasteurization. A low result confirms the product was pasteurized correctly.

How long does a test take?

About 5 minutes of incubation at 37 °C, plus about 10 seconds to read.

What equipment do I need?

A Hygiena EnSURE Touch or EnSURE luminometer, a 37 °C incubator or water bath, a 75 µL pipette and the ZymoSnap Positive Control Kit.

Is it validated?

Hygiena states ZymoSnap ALP is AOAC Performance Tested Method℠ validated. The certificate is linked above.

Can I test cream or flavoured milk?

Yes. Hygiena lists cream, flavoured milks, yogurts and shakes, as well as cow's and goat's milk. It's not suitable for UHT or other ambient-stable products.

Why do I need the positive control kit?

RLU readings depend on milk type and fat content. The positive control (350 mU/L) sets the pass/fail RLU for each product you test.

Is ZymoSnap ALP available in Canada?

Yes. Cambridge Environmental Products supplies Hygiena ZymoSnap ALP across Canada with CAD pricing.
