Hygiena InSite® L. mono Glo – Listeria spp. & L. monocytogenes Swab Test, 50 Tests

Hygiena InSite L. mono Glo is an all-in-one environmental swab test that screens for both Listeria species and Listeria monocytogenes in one device. After incubation at 37 °C, a grey or black colour shows presumptive Listeria spp. Then a green glow under a longwave UV lamp shows presumptive L. monocytogenes. Presumptive positives are visible from 24 hours and negatives are confirmed at 48 hours.

Hygiena states the test detects fewer than 10 CFU of Listeria spp. and L. monocytogenes, and holds AOAC Research Institute Performance Tested Methods℠ certification. It's sold in boxes of 50 tests. Hygiena InSite L. mono Glo is available in Canada from Cambridge Environmental Products with CAD pricing and Canadian purchasing support.

Ideal For

  • Ready-to-eat, dairy, meat and seafood plants where L. monocytogenes is the key concern
  • Environmental monitoring programs that want to separate L. mono from other Listeria species
  • Prioritizing investigations by flagging presumptive L. mono sites first
  • In-house screening before sending samples for confirmation

Why Choose InSite L. mono Glo?

Two results from one swab

Colour change screens for Listeria spp., and UV fluorescence flags presumptive L. monocytogenes in the same device. You don't need a second test or a transfer step.

All-in-one device

The pre-moistened foam swab, selective broth and indicators are in one sealed device. Hygiena states the broth includes a neutralizer to reduce sanitizer interference.

Sensitive screening

Hygiena states the test detects fewer than 10 CFU of Listeria spp. and of L. monocytogenes.

AOAC validated

Hygiena states InSite L. mono Glo holds AOAC RI Performance Tested Methods℠ certification (#061802) for L. monocytogenes and six other Listeria species on plastic, ceramic and stainless steel surfaces. The medium can also be used as a primary enrichment for Hygiena BAX® System PCR.

How to Use

  1. Swab a 10 × 10 cm (4 × 4 in) area in a crisscross pattern, rotating the swab.
  2. Return the swab to the tube, close the device and seal the joint with tape or film.
  3. Break the SnapValve and squeeze the bulb 3–4 times to release the broth.
  4. Massage the bottom of the tube and shake for 3 seconds.
  5. Incubate at 37 ± 1 °C for 24–48 hours.
  6. If the broth has turned grey or black, shine a longwave UV lamp (360–400 nm) into the side of the tube bottom and look for green fluorescence. Wear UV safety glasses.

Steps are summarized from the Hygiena InSite L. mono Glo instructions for use. Follow the full instructions.

Reading the Result

Result What You See Meaning
Negative No colour change (amber/yellow) Listeria spp. not detected (final at 48 hours)
Presumptive Listeria spp. Grey or black, no green glow under UV Presumptive Listeria spp.; confirm
Presumptive L. monocytogenes Grey or black, with green glow under UV Presumptive L. mono; confirm

Don't apply UV if the broth stays amber or yellow. A negative can only be recorded after 48 hours. Confirm presumptives with PCR, ELISA, lateral flow or a reference method such as Health Canada, FDA BAM, USDA MLG or ISO.

Selection & Handling Notes

  • UV lamp required. You need a longwave (360–400 nm) UV lamp and UV safety glasses to read the L. mono result. UV light is an eye and skin hazard.
  • Results are presumptive. Confirm positives with PCR or a reference method.
  • Wait 48 hours for negatives.
  • Listeria-like organisms such as Enterococcus can also cause blackening. Expect more presumptives from floors and drains.
  • Refrigerate on receipt. Store at 2–8 °C and use by the expiry date.
  • Trained staff only. These tests can grow pathogens. Hygiena advises that pregnant women and immunocompromised people should not be near testing.
  • Dispose safely. Treat used devices as biohazards. Autoclave, incinerate, or soak unsealed devices in 20% bleach for 1 hour before disposal.

Frequently Asked Questions

What does InSite L. mono Glo detect?

Listeria species by colour change, and presumptive L. monocytogenes by green fluorescence under UV, from the same swab.

How is it different from InSite Listeria?

InSite Listeria detects Listeria spp. only. L. mono Glo adds a UV fluorescence check that flags presumptive L. monocytogenes.

Do I need a UV lamp?

Yes. A longwave UV lamp (360–400 nm) and UV safety glasses are needed to read the L. mono result. They're not included.

How long does it take?

Presumptive positives can show from 24 hours. Negatives are recorded at 48 hours.

How should I store it?

Refrigerate at 2–8 °C and use by the expiry date.

Is InSite L. mono Glo available in Canada?

Yes. Cambridge Environmental Products supplies Hygiena InSite L. mono Glo across Canada with CAD pricing.
